Bovine oocyte maturation in vitro


Oocytes usually are cultured for 20-22 h in maturation medium at 39C, 5% CO2 in humidified air. Maturation medium is M199 with Earles salts, L-glutamine, sodium bicarbonate, and HEPES, supplemented with 7.5% (v/v) fetal calf serum, 0.5 g/ml ovine FSH, 5.0 g/ml ovine LH and 1.0 g/ml estradiol, and other small molecules that promote oocyte maturation in vitro. The cumulus cells usually expand well after maturation. Usually 75-85% oocytes are matured with first polar body extruded; indicating nuclear maturation, but cytoplasmic maturation for fertilization may be delayed.
