Ovum Pickup (OPU)


Ovum Pickup (OPU) is also called ultrasound-guided follicular aspiration. A portable ultrasound unit equipped with a 5-MHz sector scanner vaginal probe, together with a 17-gauge, 60-cm single lumen needle fitting a proper needle guide is used for trans-vaginal oocyte retrieval. Donor animals are restrained in a squeeze chute and prepared for follicular aspiration. RLI has developed the aspiration medium, Oocyte Aspiration Plus (OAP), which contains PBS based buffer medium in addition of surfactant PVA, oocyte nutrients (e.g., sodium pyruvate), antibiotics, and an anticoagulant to prevent blood clotting. Negative suction pressure was applied to maintain a low constant flow rate from 10 to 15 mL/min to collect oocytes without a disruption of cumulus cell layers. The oocytes are washed in OWP prior to IVM, and then cultured in maturation medium.
